Title: 白蝦菌株之篩選及其降解蝦殼廢棄物能力之探討

Abstract: 全世界每年約產生 6-8 萬噸蝦蟹廢棄物,若能有效利用其廢棄物,不僅可減少環境污染且可增加其經濟效益。本研究首先從白蝦 (Litopenaeus vannamei) 篩選出 135 株幾丁質分解菌 (Chitinolytic bacteria, CB)、52 株蛋白質分解菌 (Proteolytic bacteria, PB) 及 29 株乳酸菌分離菌株 (Lactic acid bacteria, LAB)。CB 菌株利用其在 Chitin plate 上透明環半徑大小選出最高之 6 株分離菌株,其中以 LV1111 菌株在 1% Chitin broth 中所得還原糖含量最高;PB 菌株利用其在 Standard methods caseinate agar 周圍之白色沉澱半徑大小選出最高之 2 株分離株 (LV1144、LV2122)。LV1144、LV2122 及 LV1111 發酵 10% 蝦殼廢棄物 (Shrimp shell waste, SSW) 所得游離胺基態氮 (Free amino nitrogen, FAN) 含量分別為 95.78、133.52 及 240.62 mg/100 mL。利用 LV2122 + LV1111混合菌株發酵 SSW 可使 FAN 提升至 295.35 mg/100 mL。LAB 菌株利用其在 MRSA + CaCO3 菌周圍產生之透明環半徑作為產酸能力之篩選,選出 3 株分離菌株 (LV3201、LV3204、LV3212),此三株菌於 10% SSW 發酵液中所得 FAN 及礦物質含量並無顯著差異,故以此三株 LAB 菌株分別與 LV1111 + LV2122 混合菌株發酵,篩選出最佳降解 SSW 之組合菌株為 CB (LV1111)、PB (LV2122) 及 LAB (LV3204)。此三株菌混合菌株發酵 SSW所得 FAN 及礦物質含量分別為單株及其他組合株發酵的 1.2-8.4 倍及 1.1-48.4 倍;總酚及總類胡蘿蔔素含量分別為 422.38 mg GAE/L 及 1.30 mg/kg,為控制組的 15.1 及 2.7倍。LV1111 + LV2122 活化菌液接種於 SSW,37oC、150 rpm 培養 2 天後再接種 LV3204 活化菌液於相同溫度下培養 3 天,所得礦物質及 FAN 含量分別為 460.0 mg/100 mL 及 18.80 mg/mL。
Shrimp and crab waste, about 6-8 million tons annually in the world, is an excellent source of biomaterials, such as chitin, protein, lipids, astaxanthin, flavor compounds, and calcium carbonate. In this study, 135 isolates of chitinolytic bacteria (CB), 52 isolates of proteolytic bacteria (PB), and 29 isolates of lactic acid bacteria (LAB) were isolated from Litopenaeus vannamei, white shrimp. 6 strains of CB were screened out by the size of clear zone on chitin plate, among them, LV1111 strain had the highest reducing sugar content in 1% chitin broth, which was 8.1 times higher than control. The content of free amino nitrogen (FAN) obtained from 10% shrimp shell waste (SSW) broth fermented by LV1144, LV2122 and LV1111 strains were 95.78, 133.52 and 240.62 mg/100 mL, respectively, and 295.35 mg/100 mL obtained from 10% SSW broth by co-fermentation of LV2122 + LV1111 strains. 3 isolates of LAB were screened out by the size of clear zone on MRSA+CaCO3. There was no significant difference in the FAN and mineral content of the 3 strains (LV3201, LV3204 and LV3212) of LAB in the 10% SSW broth. Therefore, the various combinations of LV1111, LV2122, LV3201, LV3204 and LV3212 using co-fermented technique or two-step fermented technique. The best degradation of protein and mineral on SSW two-step fermented by CB (LV1111), PB (LV2122) and LAB (LV3204).The FAN and mineral content obtained by the best combinations were 1.2-8.4 times and 1.1-48.4 times higher than individual strains and other combinations. Total phenol and carotenoid contents were 422.38 mg GAE/L and 1.30 mg/kg , which were 15.1 and 2.7 times of control. In order to reduce the cost, SSW activated broth medium was replaced by general culture medium. The FAN and mineral content obtained by LV1111 + LV2122 inoculated into SSW at 37oC, 150 rpm for 2 days, and then inoculated with LV3204 for 3 days were 460.0 mg/100 mL and 18.80 mg/mL.
