Effects of Carbohydrate Supplementation in Acute Liver Damaged Rats Induced by Carbon Tetrachloride

|Abstract: ||Abstarct:Many studies have shown that clams may improve liver function, but the mechanism is still unknown. We have reported that glycogen in clams may play an important role. To investigate the effect of carbohydrate supplementation on liver function, rats with liver damaged induced by CCI4 (1 mL/kg B.W. 20% v/ν in olive oil) was studied. Rats with liver damaged were divided into four groups: (A) control (with saline only), (B) glucose (2% w/v in saline), (c) sucrose (2% w/ν in saline), and (D) glycogen (2% w/ν in saline). 48 hours after oral administration, rats were sacrificed. Results from the acute study showed that plasma animotransferase activity and TBARS levels signifìcantly decreased after the glucose, sucrose, and glycogen treatment at the 12, 24, and 48 hour, respectively. After 48 hours of treatment, plasma bilirubin and tumor necrosis factor-α concentration significantly decreased in rats with glucose, Sucrose, or glycogen supplementation. Sucrose supplement significantly increased hepatic total glutathione, glutathione, and glycogen levels. Sucrose treatment to rats with CCI4-induced acute liver damage reduced both the blood urea nitrogen and the hepatic triglyceride content. The treatments with either glycogen or glucose given to rats decreased hepatic, TBARS levels. It is suggested that dietary carbohydrates may play a critical role in liver protection in CCI4 induced acute liver damaged rats.
摘要:許多研究指出蜆可能有改善肝功能作用，我們曾經提出報告指出蜆中的肝醣可能扮演重要的角色，為了進一步了解其機制，本研究進一步探討補充醣類對急性肝損傷大鼠之影響。本試驗使用四氯化碳來誘發大白鼠肝損傷（1mL／kg B.W.20% v/v in olive oil），肝損傷大白鼠共分成四組，分別為（Ａ）控制組（Saline only)、(B）葡萄糖組（2% w/v in saline）、（C）蔗糖組（2% w/v in saline）和（D）肝醣組（2% w/v in saline），管餵投與48小時後犧牲。試驗結果顯示，試驗期間葡萄糖、蔗糖、肝醣組都在各時間點（12,24及48小時）分別下降血漿轉胺酶活性及脂質過氧化物含量，並減少血漿脂質過氧化物、且降低血漿總膽紅索與腫瘤壞死因子TNF-α和ALT活性。急性肝損傷大鼠給予蔗糖補充，能降低血漿尿素氮濃度及肝臟三酸甘油酯之堆積，且增加肝臟總麩胱甘肽、還原態麩胱甘肽及肝醣含量。補充肝醣或葡萄糖會降低急性肝損傷大鼠肝臟脂質過氧化物。綜合以上結果，推測對於四氯化碳誘發之急性肝損傷動物模式，補充醣類可能有助於保護肝臟功能。
