Title: Detecting nitrogen deficiency with the expression of nitrate transporter gene in natural populations of the diatom genus Chaetoceros

Abstract: The nitrogen status of an ecologically important diatom genus, Chaetoceros, was estimated using the mRNA levels of nitrate transporter gene (Nrt2). Plankton samples were collected during 2 cruises to the southern East China Sea (ECS) in 2010 and 2011, and quantitative reverse-transcription polymerase chain reaction was used to measure transcript abundance of Nrt2. In addition to measuring the original expression level of Nrt2 in untreated samples, the minimum and maximum expression levels were obtained by ammonium addition and nitrogen deprivation as standards for comparison. During both cruises, low levels of Nrt2 expression were observed in Chaetoceros across the continental shelf, indicating the status of nitrogen sufficiency. The low expression levels also suggested ammonium as the major nitrogen source for Chaetocesros spp. The only high value occurred at a mid-shelf station with a deep nutricline. This expression pattern indicated that Chaetoceros spp. was not under nitrogen stress, which may be a major factor for becoming a dominant diatom group in the southern ECS.
