|Abstract: ||自基隆地區傳統市場販售的魚獲物中，隨意挑選150株產硫化氫的海洋細菌，由其中檢測出74株革蘭氏陰性長桿菌，其菌體寬度約為0.5~1.7μm，長度則為1.2~2.7μm，產硫化氫，具protease、lipase、DNase、及gelatinase等酵素活性，及極端單一鞭毛，有運動性，4℃下仍能生長，於20℃培養時可生長於含6%NaCl的培養基中，故推斷為Shewanells putrefaciens. 74株S. putrefaciens菌株經GLC生產測試，發現其細胞油脂中含 eicosapentaenoic acid (EPA)(約為總脂肪酸含量的4~5%)，且大部分的EPA存在於磷脂質中。其中以S. putrefaciens F214為分離株中EPA含量最高，菌株F214在100ml PBS broth中，於20℃振盪培養24小時後，細胞脂質中EPA的含量可達6.87%，其細胞收率為1.43g dry weight/L broth，EPA的產量為14.8mg/L broth。由以上結果可知S. putrefaciens具生產EPA之發展潛力。
Abstract: One hundred and fifty hydrogen sulfide produced marine bacteria were randomly isolated from fishery samples purchased in the fish marker of Keelung. Seventy-four of these marine isolates were obligate aerobic, gram negative, rod shaped, motile with a single polar flagellum, 0.5~1.7μm in diameter, 1.2~2.7μm in length, capable cf H2S production, and were able to grow in a medium containing 6% NaCl at 20℃. They also showed enzyme activities as protease, DNase, and gelatinase. These 74 isolates were presumely identified as Shewanella putrefaciens. As determined by GLC, all of them contained eicosapentaenoic acid (EPA) (about 4~5% of their total fatty acids). Most of the EPA were distributed in phospholipids. Isolate F214 was the highest EPA were distributed in phospholipids. Isolate F214 was the highest EPA producer among the seventy-four tested isolates. After being cultured in 100 ml PBS medium at 20℃ with shaking for 24 hours, strain F214 yielded a cell dry weight of 1.43g/L and EPA of 14.8 mg/L, which was 6.87% total fatty acid. From these results, S. putrefaciens may have potential for EPA production in the future.
