Title: 產 EPA 海洋細菌Shewanella putrefaciens 之脂肪酸組成分析

Abstract: 四種微生物油脂萃取方法中以Folch法的效果較佳,自五株Shewanella putrefaciens菌株中可萃得16.0-17.6細胞油脂。在以三種甲基酯化方法處理五株S.putrefaciens細胞油脂之脂肪酸組成氣相層析結果,經Ducan's multiple range test(P<0.05)比較後,除對脂肪酸C14:0、C18:0-C18:1與others呈現部份差異外,對於其他九種測試脂肪酸並無明顯差異,但以氫氧化鈉-鹽酸-甲醇溶液法和甲醇鈉-甲醇溶液法對於EPA的檢出有較佳的處理效果。自本省基隆地區漁獲物中篩檢出之三株產EPA S.putrefaciens菌株細胞油脂的脂肪酸組成中以棕櫚油酸(C16:1,palmitoleic acid)含量最高,佔總脂肪酸含量的27.90-28.83。而細胞油脂所含的多元不飽和脂肪酸中則以EPA為最多(為總脂肪酸含量的4.35-6.78)。
Abstract: Folch method had shown the best extraction rate, 16.0-17.6 of cellular lipid,among four bacterial lipid extraction methods used for Shewanella putrefaciens.The GC results on cellular fatty acid composition and content of S. putrefaciens derived from three methyl-esterification protocols were treated by Ducan's multiple range test (p < 0.05), and the analyzed data showed partially difference on four tested fatty acids (C14:0, C18:0, C18:1 and others). But these data were presented no difference on the remaining nine tested fatty acids, although cellularlipid of S. putrefaciens treated by NaOH-HCl-CR�記H or NaCH�烙H-CH�記Hmethyl-esterification protocol had higher content of EPA. The most abundant fatty acid in three EPA producing S. putrefaciens, which were screened frommarine fishery products purchased in the market in Keelung, was palmitoleic acid(C16:1), and its content is in the range of 27.90-28.83 of total fatty acid. EPAis the major polyunsaturated fatty acid observed (4.35-6.78 in total fatty acid).
