Title: 體型大小相似之異種異營性微細鞭毛蟲對餌料細菌攝食率差異之研究

Abstract: 本研究之異營性微細鞭毛蟲皆為分離自台灣東北沿岸之種類,依定序後得知為Placidia cafeteriopsis及Bodonid,其大小皆約為3-5 μm。為了解同樣大小不同種之異營性微細鞭毛蟲攝食反應是否有差異而進行本實驗。將兩種鞭毛蟲培養在細菌濃度9 × 106 cells mL-1 ~2.7 × 108 cells mL-1 之間。隨著餌料濃度升高,兩種之攝食率皆明顯增加P. cafeteriopsis (3.5-28.6 cells flagellate-1 h-1)、Bodonid (2.0 ~ 65.2 cells flagellate-1 h-1),但後者之變動範圍較前者大,且在一般自然水體環境濃餌料濃度條件下(<108 cells mL-1),兩者攝食速率沒有明顯差異。成長速率及半飽和常數為P. cafeteriopsis (μmax:0.12 h-1,ks:0.97 × 107 cells mL-1)、Bodonid (μmax:0.16 h-1;ks:4.08 × 107 cells mL-1)。在培養前24小時Bodonid的成長表現在體型增大,在24-36 h時其族群成長率和攝食數量有明顯正相關,而P. cafeteriopsis則延後12小時在36 h才轉變為分裂細胞以增加個體數量。由此可知大小相近但種類不同之異營性微細鞭毛蟲,其攝食成長仍存在明顯差異。
We present here the results of a study designed to analyze the potential growth and grazing activity of two different small-sized heterotrophic nanoflagellates (HNF) (2-3 μm) (Placidia cafeteriopsis vs. Bodonid) isolated from coastal surface water of northeastern Taiwan. The ingestion rate of HNF, Placidia cafeteriopsis and Bodonid, to graze on bacteria increased linearly over the whole range of bacterial density in our incubation experiment (9×106 to 2.5×108 cells mL-1). Overall, the HNF ingestion rate on bacteria for Placidia cafeteriopsis and Bodonid ranged from 3.5 to 28.6 cells flagellate-1 h-1 and 2.0 to 65.2 cells flagellate-1 h-1, respectively. Furthermore, the growth parameters (maximum growth rate and half-saturation constant) of P. cafeteriopsis and Bodonid were μmax: 0.12 h-1; ks: 9.7×106 cells mL-1 and μmax: 0.16 h-1; ks: 40.8×106 cells mL-1, respectively. Our results suggest that competitively Placidia cafeteriopsis has the advantage over Bodonid at lower bacterial abundance (<108 cells mL-1). In the first 24 hours of incubation experiment, growth in Bodonid is mainly due to increase in cell size. In the first 24-36 hours, the growth rate and ingestion rate of Bodonid are positively correlated. In P. cafeteriopsis, there is a lag of 12 hours, the number of individuals started to increase due to cell division at 36 hours after the start of incubation. It is apparent that different HNF with similar cell size may be significantly different in growth and ingestion rates.
