Title: Effects of Si-Wu-Tang In Oophorectomized Athymic BALB/c Nude Mice

Abstract: Abstract:The aim of this study was to investigate the effects of Si-Wu-Tang (SWT) on breast tumor growth and uterus weight in athymic BALB/c nude mice. Human breast cancer MCF-7 cells were implanted in subcutaneous sites of oophorectomized mice. Three days after bilateral oophorectomy, mice were randomly separated into 5 groups: vehicle, 0.3 x SWT (equivalent to 25.8 mg SWT herb/20 g mouse/day), 1 x SWT, 3 x SWT and 1 positive control (11 μmole of 17β-estradiol, E2). Mice were administrated SWT extracts via feeding tube at every morning. Positive control mice were subcutaneous injection E2 every 3 days. The effects of SWT on tumor growth and uterus weight were assessed after 21 days of treatment. The H&E stain and microscopy examinations were preformed to measure the thickness of dermal layer and the present of tumor foci. The results showed that subcutaneous injection of E2 significantly increased uterus weight to 4 folds of the control group. Skin lesions were noticed on 75% of 1 x SWT group and 50% of E2 group, respectively. The thickness of dermal layer was increased in 1 x SWT group. In addition, hyalinization degenerations under dermal layer were also observed in 3 x SWT and E2 group. Furthermore, tumor cells were presented in E2 group. These findings suggested that the duration of treatment and dosage of SWT need to be modified. Besides, the pharmacokinetics studies of SWT in oophorectomized athymic BALB/c nude mice need to be investigated.
