Title: 台灣鮑萎縮症流行病學調查研究
Study on the pathology of atrophy in Taiwan small abalone (Haliotis diversicolor supertexta)

Abstract: 本研究以2007 ~ 2009年間發生萎縮症的病鮑作為實驗樣本,分別自台灣宜蘭頭城及竹安地區的鮑養殖場作長期採樣。採集的樣本,經初步觀察其體表有明顯的萎縮症狀,同時有軟組織萎縮 (包含肌肉和內臟團) 、不進食、活動力弱、翻面困難、足部黑變及敏感度差等狀況,嚴重者則會出現軟組織萎縮後,大量死亡的現象。 由養殖場取得萎縮病徵的病鮑,測量計算軟組織重/體全重、軟組織重/殼重等比例,與健康的鮑體比較分析在軟組織的萎縮程度有極大的差異;而在飢餓實驗結果分析顯示,病鮑所呈現之萎縮症典型病癥應為罹病所致,而非因飢餓所造成的。在組織切片病理觀察方面,結果顯示足部肌肉、腸道、鰓與生殖巢等這些部位組織均有明顯的病徵;病理組及攻擊組的組織細胞較健康組有明顯組織異常的現象。 利用聚合酶連鎖反應檢測Rickettsiales-like prokaryote (RLP) ,以27F 與 1525R之引子由編號070801、070831、071026-11、080610、081110、090326樣本中選殖到1455 bp 的PCR產物,而其基因序列與類立克次體 (AF133090) 有78 % 的相似度。除此之外,以C6F/C1R、C6F/C2R、OsR/OsF之皰疹病毒檢測引子,並未選殖到Herpes-like virus。 以上結果推測發生萎縮症之病鮑其病原可能為類立克次體,但可能因為地理環境上的不同而在基因型上有些微差異。
Withering Syndrome (WS) is a chronic and serious syndrome caused by infectious agent in abalone (Haliotis spp.), and the syndrome broke out since 2002 in Taiwan. In this study, the detection and pathology of Withering Syndrome (WS) from culture small abalone in coast of North-eastern Taiwan was investigated during 2008 of 2009. The symptom of WS observed was similar to that of starvation, exhibiting weakness, poor activity, anorexia, body mass loss and death in small abalone. The histopathological results showed atrophy in myofibers of the foot muscle and the broke out and solved of digestive gland tissue. After challenge experiments the injected abalone showed similar abnormal symptoms in foot muscle, digestive gland tissue, gill and gonad. The pathological features of atrophy in infected abalone showed differences in soft tissue weight / total body weight, soft tissue weight / shell weight comparing with that of healthy abalone. Using the polymerase chain reaction (PCR) technology to detect of Rickettsia-like prokaryote (RLP) with the pair of primers 27F/1525R, an 1455 bp PCR product was detected in samples 070801, 070831, 071026-11, 080610, 081110 and 090326, which was highly similar with Rickettsia-like (AF133090) with a similarity of 78 %. However, no PCR product was cloned from the samples using pairs of primers C6F/C1R, C6F/C2R, and OsF/OsR to detect Herpes-like virus. The present result indicate that the withering syndrome of abalone is caused by the infection of a Rickettsia-like prokaryote, which the genetic differences maybe due to the variation of host and geography.
