Title: 嗜水性產氣單胞菌疫苗應用於養殖吳郭魚之研究

Abstract: 本研究利用常發生於淡水魚之嗜水性產氣單胞菌Aeromonas hydrophila strain AAH,以不同培養基製備成菌苗,經免疫處理吳郭魚後進行保護效果探討。主要內容包括不同培養基對於 A. hydrophila 細胞外產物之溶血活性與蛋白質分解酵素活性影響、吳郭魚保護效果之 毒性試驗及血清之抗體反應試驗。 實驗結果發現,在不同培養基對於 A. hydrophila 之細胞外產物影響方面,培養基TSA + 微量元素(O. salts)+ 2% Glucose + 鐵螯合劑(100 μM 2, 2-dipyridyl),具有增強蛋白質分解酵素活性及增強溶血活性的效果。 免疫保護效果試驗,將 A. hydrophila strain AAH以五組不同的培養基製備不活化菌苗,以腹腔注射方式施打吳郭魚亞成魚。結果以TSA + O. salts + 2 % Glucose 組菌苗,第四週活存率 80 % 具保護效果,而 以 TSA + O. salts + 2 % Glucose + 100 μM 2, 2-dipyridyl 組菌苗,第四週活存率 70 %,亦具保護效果,但在第八週後菌苗保護效果降低活存率僅剩 20 – 40 %。而將菌苗以浸泡的方式施予,似乎無法提供有效的保護效果。因此推論本試驗以TSA + O. salts + 2 % Glucose 與 TSA + O. salts + 2 % Glucose + 100 μM 2, 2-dipyridyl 為培養基製作菌苗,對吳 郭魚僅具有短期的保護效果。
This study investigated the efficacy of Aeromonas hydrophila vaccines prepared from different culture media, in tilapia. The main content included the detection of protease activity and haemolytic units variation of ECP of Aeromonas hydrophila grown on different media, survival rates of tilapia preimmunized with various vaccine, rocket immuno-electrophoresis and agglutinating activity of tilapia antisera. The medium, TSA + O.salts + 2 % glucose + 100 μM 2, 2-dipyridyl, enhanced higher production of protease and haemolytic activities than other media. In immunological protection tests, TSA + O.salts + 2 % glucose group vaccine resulted in 80 percent survival rate while TSA + O. salts + 2 % glucose of + 100 μM 2, 2- dipyridyl group vaccine resulted in 70 percent survival rate for four weeks. But the protection reduced to only 20-40 percent after eight week for all the vaccine groups. And the vaccines given to fish by way of immersion resulted in no protection. It seems that the immunization using both TSA + O.salts + 2 % glucose group and TSA + O.salts + 2 % glucose of + 100μM 2, 2- dipyridyl group vaccines only result in a short-term protection in the tilapia.
