Title: 龍鬚菜與石花菜熱水萃取物強化白蝦免疫力及抵抗Vibrio alginolyticus感染之研究

Abstract: 摘要 白蝦(Litopenaeus vannamei)經龍鬚菜(Gracilaria tenuistipitata)或石花菜(Gelidium amansii)之熱水萃取物注射處理後,研究其對白蝦非特異性免疫因子之影響,以及其對於抗Vibrio alginolyticus之抵抗力。 白蝦經注射4 μg g-1及6 μg g-1之龍鬚菜熱水萃取物,經第1及2天後,其總血球數、酚氧化酵素活性及超氧離子均顯著增加。超氧歧化酵素活性於注射後第1天顯著增加。白蝦注射6 μg g-1之龍鬚菜熱水萃取物後,感染V. alginolyticus(1×107 cfu shrimp-1),1天後,其吞噬作用及清除能力明顯增加。白蝦注射6 μg g-1龍鬚菜熱水萃取物後,感染V. alginolyticus(2×106 cfu shrimp-1),然後放置於原來34 ‰ 之海水中,經3天後,其存活率明顯高於注射生理食鹽水組及對照組。證實白蝦經龍鬚菜熱水萃取物處理後能增加其免疫力及抵抗V. alginolyticus之感染。 白蝦經注射6 μg g-1石花菜熱水萃取物1天後,其總血球數、酚氧化酵素活性及超氧離子明顯高於對照組。2天後,其吞噬作用及清除能力明顯增加。白蝦注射6 μg g-1石花菜熱水萃取後,感染V. alginolyticus(2×106 cfu shrimp-1), 2天後,其活存率明顯高於生理食鹽水組及對照組,且持續至6天後其存活率仍明顯高於對照組。顯示白蝦經石花菜熱水萃取物處理後能增加其非特異性免疫力及抵抗V. alginolyticus之感染。 本研究證實白蝦經龍鬚菜或石花菜熱水萃取物注射後,具有增強白蝦免疫力,且增強其抵抗V. alginolyticus感染。其中又以注射龍鬚菜熱水萃取物之效果較好,但持續的添加是有必要的。 關鍵字:白蝦、龍鬚菜、石花菜、熱水萃取物、Vibrio alginolyticus、免疫因子、感染實驗、吞噬作用、清除能力。
Abstract This study examined the immune response of the white shrimp Litopenaeus vannamei (8.4∼12.4 g) and its resistance against Vibrio alginolyticus when the shrimp were injected with hot-water extract of Gracilaria tenuistipitata or Gelidium amansii at a dose of 4 or 6 μg g-1. The immune parameters included total haemocyte count (THC), phenoloxidase activity, respiratory burst (release of superoxide anion), superoxide dismutase activity, phagocytic activity and clearance efficiency. L. vannamei injected with hot-water extract of G. tenuistipitata at 6 μg g-1 increased its THC、phenoloxidase activity and respiratory burst after 1 and 2 days. L. vannamei injected with hot-water extract of G. tenuistipitata at 6 μg g-1 increased its superoxide dismutase activity after 1 day. L. vannamei injected with hot-water extract of G. tenuistipitata at 6 μg g-1 increased its phagocytic activity and clearance efficiency to V. alginolyticus after 1 day. In another experiment, L.vannamei which had been injected with hot-water extract of G. tenuistipitata, were challenged with V. alginolyticus at 2×106 colony-forming units (cfu) shrimp-1 and then placed in seawater of 34 ‰. The survival of shrimp that received hot-water extract of G. tenuistipitata at 6 μg g-1 was significantly higher than that of shrimp injected saline and the control shrimp at the termination of the experiment (6 days after the challenge). It is concluded that hot-water extract of G. tenuistipitata at a dose of 6 μg g-1 causes an increases in the immune ability of L. vannamei and resistance from V. alginolyticus infection. L. vannamei injected with hot-water extract of G. amansii at 6 μg g-1 increased its THC、phenoloxidase activity and respiratory burst after 1 day. No significant difference in superoxide dismutase activity was observed among the shrimp injected with saline and those injected with hot-water extract of G. amansii at doses of 4 and 6 μg g-1. L. vannamei injected with hot-water extract of G. amansii at 6 μg g-1 increased its phagocytic activity and clearance efficiency against V. alginolyticus after 2 days. In another experiment, L. vannamei which had been injected with hot-water extract of G. amansii were challenged with V. alginolyticus at 2×106 cfu shrimp-1, and then placed in seawater of 34 ‰. The survival of shrimp that received hot-water extract of G. amansii at 6 μg g-1 was significantly higher than that of shrimp injected saline and the control shrimp after 6 days. The present study indicated that both hot-water extracts of G. tenuistipitata and G. amansii can enhance the immune ability of L. vannamei. However a continuous dose is necessary to maintain the immune ability of shrimp. Further research is needed to evaluate the immunostimulatory effects of L. vannamei and its resistance against V. alginolyticus when shrimp fed a diet containing either hot-water extract of G. tenuistipitata or G. amansii .
