Title: 台灣東北部沿岸Spirotontonia taiwanica n. sp.形態與小次單位核醣體DNA序列比對之研究

Abstract: 依據蛋白銀染色法所得之形態特徵與18S rRNA之序列判定Spirotontonia taiwanica n. sp.為一新種;本研究提供本種18S rRNA之序列資料為Tontoniidae科第一個登入之基因序列。 Spirotontonia taiwanica之特徵簡介如下:使用蛋白銀染色後蟲體長寬分別為92 μm (80-104 μm)及53 μm (40-72 μm)。大核近圓形 (6.8 × 6.6 μm),數目多達25顆 (20 - 31)。口圍帶由13 (12 - 15)片領區小膜及12 (11 - 13)片口區小膜組成。蟲體之伸縮性尾部位於背側末端第一列與第二列腰動基列間,尾部表面動基粒沿著一定方向呈現不規則之排列,據此判定此新種屬於Tontoniidae科。腰動基列由口區小膜末端向左環繞蟲體2.5圈至蟲體末端,據此判定此種屬於Spirotontonia屬。此屬加上本研究種目前全球僅有三種,本新種因沒有皮質小板及腰動基列之圈數較多,可與其他兩種分辨。 演化樹結果得知,本新種位於少毛目之群集內,並與Laboea strobila親源關係最為相近。証明纖毛圖示在演化上較尾部佔有更重要之演化地位。
A new species Spirotontonia taiwanica n. sp. was established according to morphological data from protargol impregnation and 18S rRNA genetic data, which was the first registered data of Tontoniidae in the GenBank/EMBL databases. The size of Spirotontonia taiwanica 92×53 μm (80-104×40-72 μm) after protargol impregnation. Macronuclear nodules numerous, about 25 (20-31), size 6.8×6.6 μm, oval-shaped. Adoral zone of membranelles composed of 12-15 collar membranelles and 11-13 buccal membranelles. Contractile tail inserts between first and second whorl of girdle kinety on dorsal side. As a result, we determined this species to Tontoniidae. Furthermore, girdle kinety sinistrally spiraled ~2.5 circle, composed of dikinetids. According to this feature, we determined this species to Spirotontonia. However, There are only three species including S. taiwanica in Spirotontonia in the whole world. The new species was different from other species because of the absence of cortical plates and the number of spiralled circles of girdle kinety. According to the result of the phylogenetic tree, the new species clustered with Laboea strobila and group within the order Oligotrichida. We proved the character of infraciliature was more important than that of tail in the course of evolution.
